(pro)-OpenCRISPR-1-SV40pA + U6-(gRNA)-CAG-mCherry-bGHpA

OpenCRISPR-1 counterpart to the SpCas9 single-gRNA CAG-mCherry design. Nuclease AAV expresses AI-designed OpenCRISPR-1 under a custom promoter with SV40pA; companion AAV delivers one U6-driven gRNA and CAG-mCherry for broad whole-cell fluorescent verification of transduced cells. Canonical SpCas9 sgRNAs are fully compatible.

Characteristics

OpenCRISPR-1 nuclease with 95% off-target reduction and reduced immunogenicity vs SpCas9. CAG-mCherry provides ubiquitous red-channel whole-cell fluorescence without nuclear membrane targeting. Green channel is free for co-reporters or imaging probes. Compact payload — no KASH domain.

Applications: Single-target editing where whole-cell red fluorescence verification is sufficient and FANS is not required. Preferred when green channel must remain free. Drop-in replacement for SpCas9 counterpart using existing gRNA designs.
