CMV early enhancer/chicken beta-actin (CAG) promoter

Synthetic hybrid promoter composed of the CMV immediate-early enhancer and chicken β-actin promoter, enabling strong, ubiquitous expression across most mammalian cell types. Produces very high initial expression but may undergo silencing in certain tissues over long-term in vivo studies.

Characteristics

Strong ubiquitous constitutive mammalian promoter (~1.7 kb); CMV IE enhancer + chicken β-actin promoter + rabbit β-globin intron. Niwa et al. 1991. Standard for high-level transgene expression.

Applications: When maximum expression is needed across diverse cell types. Popular for transgenic animals and stable cell lines. Less compact than EFS.
