U6-(gRNA)-hSyn-EGFP-KASH-hGHpA

Single-gRNA vector expressing EGFP-KASH from the neuron-specific hSyn promoter for FANS-compatible nuclear envelope labeling of transduced neurons. Derived from the Platt et al. 2014 hSyn-EGFP-KASH architecture (Addgene #60231) with Cre and WPRE removed and hGHpA as terminator. Smaller and simpler than the Cre-containing parent for use cases where recombinase activity is not required.

Characteristics

Compact single-cassette design. hSyn restricts EGFP-KASH expression to neurons in mixed-cell tissue. EGFP-KASH localizes to the outer nuclear membrane enabling FANS-based isolation of transduced neuronal nuclei. Removal of WPRE and Cre reduces payload size relative to #60231, extending usable insert capacity.

Applications: Companion gRNA vector for in vivo CRISPR editing in neurons where Cre activity is not needed. EGFP-KASH enables FANS-based nuclei sorting from brain tissue for downstream snRNA-seq or snATAC-seq. Green channel preserved for co-imaging; pair with mCherry-KASH variants when green channel must remain free.
