- LbCas12a-RVRR Nuclease: PAM-altered variant of Lachnospiraceae bacterium Cas12a (LbCas12a) carrying four substitutions that broaden the enzyme's protospacer adjacent motif requirement from the canonical TTTV to a TNTN consensus. Expands the range of genomic sites accessible to Cas12a-based genome editing and cytidine base editing beyond the wild-type PAM constraint.
- DNMT3A Domain: Targeted epigenetic silencing through DNA methylation. Creates stable, heritable gene repression. Particularly effective at CpG islands. Methylation persists through cell divisions. Larger than transcriptional repressors (~300 amino acids for catalytic domain). Slow-acting but durable.
- VP64: Synthetic transcriptional activator comprising four tandem repeats of the VP16 minimal activation domain. Engineered by Beerli et al. for enhanced gene activation, achieving 27-fold transcriptional upregulation versus 5-fold with native VP16 when fused to zinc finger proteins.
- Rta Domain: Component of VPR tripartite activator. Enhances activation when combined with VP64 and p65. Adds minimal size (~100 amino acids) but significant functional benefit. Particularly useful for activating viral promoters and some cellular genes.
- p65 Domain: Strong transcriptional activator used in combination systems (e.g., VPR). More potent than VP64 alone but larger. Commonly combined with VP64 and Rta for enhanced activation. Particularly effective for difficult-to-activate genes.
Specifications
Single-stranded AAV produced in HEK293T cells via triple transfection and purified by two rounds of CsCl ultracentrifugation. Capsid serotype, promoter, and transgene configured per order. Titer determined by qPCR targeting common and customer-specified elements. Full capsid content ≥85% by mass photometry. Endotoxin ≤1 EU per 1×10¹³ VG. Purity ≥95% by SDS-PAGE. Formulated in DPBS supplemented with 150 mM NaCl and 0.001% Pluronic F-68. Delivered with batch report and payload sequence file. Store at −80°C long-term. Stable at −20°C for up to 2 months and at 4°C for 1–2 weeks after thaw.
