(pro)-OpenCRISPR-1-SV40pA + U6-(gRNA)-(ins)

OpenCRISPR-1 counterpart to the minimal SpCas9 single-gRNA custom-insert design. Nuclease AAV expresses AI-designed OpenCRISPR-1 under a custom promoter with SV40pA; companion AAV delivers one U6-driven gRNA alongside a user-defined insert slot. No fluorescent reporter — maximum payload capacity available for the custom insert. Canonical SpCas9 sgRNAs are fully compatible.

Characteristics

Most payload-efficient OpenCRISPR-1 dual-AAV configuration. OpenCRISPR-1 nuclease at 4.1 kb is the same size as SpCas9, preserving the same payload budget on the nuclease AAV. Absence of reporter maximizes insert capacity on the companion AAV. Off-target profile (95% reduction vs SpCas9) is particularly relevant for HDR template co-delivery where precision is critical.

Validated in: HEK293 cells (OpenCRISPR-1 characterization)

Applications: Single-target editing requiring co-delivery of a custom payload (HDR donor, transgene, or additional expression element) with no reporter overhead. Preferred over SpCas9 counterpart when the insert is an HDR template or therapeutic cargo where off-target cleavage would be most consequential.
