Architecture

Catalytically dead Cas9 fused to catalytic domain of ten-eleven translocation 1 (TET1) enzyme. Converts 5-methylcytosine to 5-hydroxymethylcytosine, initiating demethylation pathway.

Characteristics

Targeted DNA demethylation system for epigenetic activation. Removes methylation marks at target loci to reactivate silenced genes. Effective at reversing promoter methylation. Can restore expression of epigenetically silenced tumor suppressors or developmental genes.
