VPR Composite Domain

Synthetic tripartite transcriptional activator fusing VP64, p65, and Rta to drive 10–1000× gene upregulation in CRISPRa systems. Synergistic activity from three complementary activation mechanisms exceeds any individual domain. Gold-standard effector domain for activating silenced, low-expression, or refractory genomic loci where VP64 alone is insufficient.

Mechanism

Tripartite activation domain combining three complementary activation mechanisms. VP64 provides core activation, p65 enhances chromatin remodeling, Rta adds viral transactivation function. Synergistic effect exceeds individual domains.

Characteristics

Enhanced CRISPRa system with very strong activation (10-1000× typical). Larger than VP64 alone (~250 amino acids total). Gold standard for difficult-to-activate genes. May cause some cellular stress at very high expression levels. Most publications use VPR for maximum activation.
