Mechanism

Repeating peptide epitopes (typically 10-24 copies) recognized by single-chain antibody fragments (scFv). Each GCN4 epitope recruits one scFv-effector fusion, amplifying effector domains at target site. Scaffold strategy for signal amplification.

Characteristics

Peptide array for recruiting multiple effector domains per dCas9 (SunTag system). Typically 10× amplification of VP64 or other effectors. Requires two-component system (dCas9-GCN4 + scFv-effector). Very strong activation but complex. Large total size due to repeats. Most potent activation system available.
