Two-vector system with SaCBE N-terminal and KKH-enhanced C-terminal for expanded NNNRRT PAM targeting

Characteristics

First AAV delivers SaCBE N-terminal (APOBEC1 + SaCas9 aa1-534) under EFS promoter. Second AAV delivers SaCBE-KKH C-terminal (SaCas9 aa535-1053 with E782K/N968K/R1015H mutations) under selected promoter with U6-gRNA. KKH mutations expand PAM from NNGRRT to NNNRRT. Split at Lys534/Cys535 using Npu DnaE intein. Reconstitutes full SaCBE-KKH activity in target cells.
