Characteristics

First AAV delivers SaCBE N-terminal (APOBEC1 + SaCas9 aa1-534) under EFS promoter. Second AAV delivers SaCBE C-terminal (SaCas9 aa535-1053) under selected promoter with U6-gRNA. Split at Lys534/Cys535 using Npu DnaE intein. Reconstitutes full SaCBE activity in target cells. NNGRRT PAM recognition.
