Characteristics
First AAV delivers SaBE3.9max N-terminal (codon-optimized APOBEC1 + enhanced NLS + SaCas9 aa1-534) under EFS promoter. Second AAV delivers SaBE3.9max-KKH C-terminal (SaCas9 aa535-1053 with E782K/N968K/R1015H mutations) under selected promoter with U6-gRNA. Combines optimization with PAM expansion (NNNRRT). Split at Lys534/Cys535 using Npu DnaE intein. Highest activity SaCas9 CBE variant with broadest PAM compatibility.
