Characteristics
First AAV delivers SaBE3.9max N-terminal (codon-optimized APOBEC1 + enhanced NLS + SaCas9 aa1-534) under EFS promoter. Second AAV delivers SaBE3.9max C-terminal (SaCas9 aa535-1053) under selected promoter with U6-gRNA. Split at Lys534/Cys535 using Npu DnaE intein. Reconstitutes full SaBE3.9max activity with improved expression and efficiency. NNGRRT PAM recognition.
