psPAX2

HIV-1 packaging plasmid for lentiviral vector production: a CAG promoter (CMV enhancer, chicken β-actin promoter and a chimeric intron) drives HIV-1 gag and pol, with the cPPT/CTS and the RRE, followed by a rabbit β-globin polyadenylation signal. Deposited by Didier Trono (EPFL) as Addgene #12260, whose description calls it a 2nd generation lentiviral packaging plasmid; it supplies the packaging proteins in trans to a transfer vector and an envelope plasmid.

Characteristics

10,711 bp circular plasmid with ampicillin resistance in bacteria. The file annotates the CMV enhancer, chicken β-actin promoter and chimeric intron, HIV-1 gag (1,503 bp) and pol (3,012 bp, overlapping gag by 208 bp), the cPPT/CTS, the RRE (234 bp), a gp41 epitope peptide, an HIV Rev nuclear export signal, a rabbit β-globin poly(A) signal, and an SV40 promoter, origin and poly(A) signal, then a pUC-type origin and AmpR. It annotates no long terminal repeats, packaging signal or envelope gene, so it is not itself packageable. The Addgene description line in the file is cut off after 'Can be used with 2nd or'. Addgene lists the plasmid as unpublished, a Trono lab packaging plasmid with no describing paper. A citing paper derived an integrase-deficient (D64E) packaging cassette from it.

Additional information

Applications: Supplying HIV-1 Gag and Gag-Pol packaging functions in trans for lentiviral vector production, by co-transfection of producer cells such as HEK293T with a lentiviral transfer vector and an envelope plasmid (commonly VSV-G). Also used as the parent of integrase-deficient packaging constructs.
