Compact Staphylococcus aureus Cas9 nuclease (3.2 kb) that fits alongside a guide RNA and regulatory elements within a single AAV vector. Requires NNGRRT PAM, with the KKH variant expanding recognition to NNNRRT for broader genomic coverage. First validated compact alternative to SpCas9 for in vivo gene editing in size-constrained therapeutic vectors.
