U6-(gRNA)-(pro)-Target-AID-bGHpA

Express gRNA under U6 and Target-AID under selected pro for narrow-window cytosine editing with reduced bystander mutations

Characteristics

Uses PmCDA1 (activation-induced cytidine deaminase from sea lamprey) with narrower editing window at positions 2-4, reducing bystander C-to-T conversions. Lower off-target activity compared to APOBEC1-based editors. NGG PAM with SpCas9 D10A nickase. Requires dual-AAV packaging. Preferred when precise positional control is needed.

Custom Inserts

SpCas9 gRNA target (20-23 bp)

Additional Information

Applications: Targets with multiple cytosines where bystander editing would be deleterious. Applications requiring editing at specific position with minimal adjacent C conversion. When narrow editing window provides selectivity advantage. Precision therapeutics where unintended C-to-T conversions could create new pathogenic variants.
