U6-(gRNA)-(pro)-SaABE8e-V106W-bGHpA
Express sgRNA under U6 and SaABE8e V106W under selected promoter for minimized off-target RNA editing
Characteristics
V106W mutation in TadA* domain reduces off-target RNA editing 7.2-fold (from 0.49% to 0.068% average frequency) while maintaining robust DNA editing. Provides 2.7-fold improvement in off-target DNA editing specificity and 3.7-fold reduction in indel formation compared to parent SaABE8e. NNGRRT PAM recognition.

Additional Information
Applications: Therapeutic development requiring minimal transcriptome perturbation and highest specificity. Long-term expression applications where cumulative RNA editing is a concern. When target validation or clinical translation demands lowest possible off-target effects across DNA and RNA.
