pCMV-GFP

Mammalian expression plasmid in which a CMV enhancer and promoter drives EGFP, followed by a rabbit β-globin polyadenylation signal. Deposited by the Cepko lab as Addgene #11153 on a pCAGEN backbone; Matsuda and Cepko made it by replacing the CAG promoter of pCAG-GFP with the CMV promoter from pEGFP-N1.

Characteristics

4,479 bp circular plasmid with ampicillin resistance in bacteria. In the developing rat retina, GFP from the CMV promoter was lower than from the EF1α or ubiquitin C promoters, and it labeled cells with bodies in the inner nuclear layer more than photoreceptors, suggesting weak activity in photoreceptors.

Additional information

Applications: Expressing GFP from the CMV promoter in mammalian cells; in the retina work that introduced it, CMV was a poor choice when photoreceptors need to be labeled.
