AAV5 Serotype
Natural serotype with unique receptor usage and distinct tropism profile. Notable for lower pre-existing immunity and efficient CNS transduction.
Characteristics
Uses platelet-derived growth factor receptor (PDGFR) and sialic acid (N-linked glycans) as receptors, not HSPG. Lower pre-existing immunity versus AAV2 (~40% seropositive). Efficient airway epithelial transduction from apical surface. Strong photoreceptor tropism in retina. Moderate liver transduction. Slower intracellular trafficking.
Applications
Retinal gene therapy (alternative to AAV2). Cystic fibrosis and lung disease gene therapy. CNS applications. Photoreceptor-targeted treatments. Applications in antibody-positive patients.
