Natural serotype with unique receptor usage and distinct tropism profile. Notable for lower pre-existing immunity and efficient CNS transduction.

Characteristics

Uses platelet-derived growth factor receptor (PDGFR) and sialic acid (N-linked glycans) as receptors, not HSPG. Lower pre-existing immunity versus AAV2 (~40% seropositive). Efficient airway epithelial transduction from apical surface. Strong photoreceptor tropism in retina. Moderate liver transduction. Slower intracellular trafficking.

Applications: Retinal gene therapy (alternative to AAV2). Cystic fibrosis and lung disease gene therapy. CNS applications. Photoreceptor-targeted treatments. Applications in antibody-positive patients.
