Rhesus-derived serotype with broad tissue tropism and notably lower human seroprevalence. Clinical relevance for patients with pre-existing AAV immunity. Template for Spark100 engineering.
Characteristics
Isolated from rhesus macaque tissue. Very low pre-existing neutralizing antibodies in humans (~5-15% seropositive). Broad tissue tropism across liver, muscle, and CNS. Good transduction efficiency. Favorable immune profile. Template for Spark100 clinical vector. Strong safety profile in preclinical studies.

Gene therapy in antibody-positive patients. Hemophilia trials (Spark Therapeutics). Liver-directed metabolic disorders. CNS applications. Re-administration protocols. Template for Spark100 development (hemophilia B clinical success).
